Vaccinia virus A27L-3stop
This part codes A27L-3stop, which is derived from the A27L mutation, where the codons of three phenylalanines at the N-terminus are changed to the amber suppressor codon UAG. Only when a specific artificial translation system is introduced and the corresponding unnatural amino acids are added (see BBa_K5142006), the modified vaccinia virus with complete A27L protein can be synthesized. Under normal conditions, the A27L protein cannot be fully synthesized, and thus EEV production is significantly reduced, so that most viruses remain in the cells as IMVs, thereby the safety regarding using the virus can be enhanced.
This part is derived from the vaccinia virus A27L (BBa_K5142006) where the codons of three phenylalanines at the N-terminus are changed to the amber suppressor codon AUG.
According to the three-dimensional structure model of the protein, the N-terminus of A27L is located on the surface of the viral particle. After incorporating 4-azido-L-phenylalanine, the azide group can be link to customized ligands by click reaction of azido and dibenzocyclooctyne (DBCO). Please see BBa_K5142006 for more details.
